Velocity of Sarcomere Shortening in Rat Cardiac Trabeculae
نویسنده
چکیده
Several recent reports have indicated that catecholamines may act directly on the crossbridge cycle, independent of intracellular calcium concentration changes. The present study investigated the effect of isoproterenol on peak force during twitches at constant sarcomere length and unloaded velocity of sarcomere shortening in isolated right ventricular trabeculae of hearts with V, or V3 isomyosin obtained from euthyroid and hypothyroid rats, respectively. Hypothyroidism was induced by treatment of the rats with propylthiouracil for 6 weeks. Electrophoretic analysis showed that the hearts of hypothyroid animals were composed only ofV3 isomyosin, whereas the hearts of euthyroid animals were composed predominantly of V1 isomyosin. Force development was measured with a silicon strain gauge and sarcomere length with laser diffraction techniques; the shortening velocity was determined from contractions in which sarcomere length was initially held constant followed by a quick release to zero load and a controlled release at zero load. Both isometric twitch force and unloaded sarcomere shortening velocity

Whether β-adrenergic stimulation affects the cross-bridge cycling rate independently of its effect on Ca2+ handling by the cardiac myocyte is still unknown. An increase in cross-bridge cycling rate may result in increased unloaded velocity of sarcomere shortening ( V o). To test this hypothesis directly, skinned rat cardiac trabeculae were attached between a silicon strain gauge (∼3.5 kHz reson...

Sarcomere length measurement by microscopic and laser diffraction techniques in trabeculae of rat heart, superfused with Krebs-Henseleit solution at 21 degrees C, showed spontaneous local sarcomere shortening after electrically stimulated twitches. The contractions originated in a region of several hundred micrometers throughout the width of the muscle close to the end of the preparation that w...

The effect of temperature on the force-sarcomere velocity relation (200, 250, and 300 C) and maximum velocity of sarcomere shortening (V0; range 15°-35° C) was studied in trabeculae from rat heart. Sarcomere length and V. were measured by laser diffraction techniques. Sarcomere length and sarcomere velocity, determined from each of the first-order diffraction lines, differed by less than 4%. Sl...
متن کاملThe force-velocity relation and stepwise shortening in cardiac muscle.
A series of experiments was carried out to determine the effects of load variation on the character of stepwise shortening. We imposed afterloaded isotonic contractions on rat ventricular trabeculae, and measured the effect of load on pause duration, i.e., on the duration of the periods during which there was no sarcomere shortening. Sarcomere lengths were measured by optical diffraction. Incre...
